Analysis of Tritium Incorporation into Individual Cells by Autoradiography of Squash Preparations
نویسندگان
چکیده
The relation between tritium content of individual cells and grain count obtained in autoradiographs of squashed cells was investigated. The tissues used were root meristems of Tradescantia paludosa and intestinal epithelium of the mouse. The relation between grain count and tritium content is affected by self-absorption which depends on the thickness of the labeled cell. Therefore, squashed preparations were sectioned to determine the uniformity of thickness of nuclei. In a preparation of mouse cells, thicknesses were 1.18 +/- 0.35 micro, and in a preparation of Tradescantia cells, 2.97 +/- 0.35 micro. The effects of similar and larger variations in thickness upon grain count were studied in material squashed with different pressures; no marked correlation was found. The lack of correlation is explained by the geometric relation between labeled nuclei and the emulsion. By counting grains and directly measuring tritium content in a glass proportional counting tube in the same preparation, the yield of grains per disintegration was measured in Tradescantia cells and found to be 1 grain for 10.9 disintegrations with AR 10 autoradiographic film and 1 grain for 19.3 disintegrations for NTB nuclear track liquid emulsion. Latent image fading may pose a problem with long exposures; the conditions of its occurrence are as yet not well known.

Formation and detection of RNA-DNA hybrid molecules in cytological preparations.
A technique is described for forming molecular hybrids between RNA in solution and the DNA of intact cytological preparations. Cells in a conventional tissue squash are immobilized under a thin layer of agar. Next they are treated with alkali to denature the DNA and then incubated with tritium-labeled RNA. The hybrids are detected by autoradiography. The technique is illustrated by the hybridiz...
متن کاملStudies on tritiated thymidine incorporation into DNA molecules by electron microscopic autoradiography.
The incorporation of tritiated thymidine into DNA molecules was studied by electr.:m microscopic autoradiography. To make autoradiogram in electron microscopic level, DNA was extracted from rat ascites hepatoma (AH 130) cells after in vitro incubation with tritiated thymidine. Extracted DNA samples were rotary shadowed with platinum palladium and covered with emulsion. Silver grains demonstrate...
متن کاملDiagnostic Accuracy of Squash Preparations in Central Nervous System Tumors
Background & Objectives: The accurate assessment of the diseased tissue is fundamental to the diagnosis and management of disorders of the central nervous system (CNS). The ‛squash’ or ‛crush’ technique has been universally employed in the intraoperative diagnosis of CNS tumors. The aim of our study was to evaluate the accuracy of squash preparation in diagnosing CNS tumors by comparing with hi...
متن کاملIncorporation of a potent antileukemic agent, 5-aza-2'-deoxycytidine, into DNA of cells from leukemic mice.
5-Aza-2'-deoxycytidine administered at a daily dose of 1.5 mg/kg increased the life-span of P388 leukemia-bearing BALB/c X DBA/2 F1 mice by 5 times and that of second generation lymphoma-bearing AKR mice by 2.5 times. Higher doses (total dose, 20 mg/kg) led to favorable results when administered in two portions on Days 4 and 5 after the s.c. inoculation of leukemic cells. The same total dose gi...
متن کاملEvidence for Cytoplasmic Dna in Root Cells of Nicotiana
Sterile root cultures from Nicotiana tabacum were grown with H(3)-thymidine added to the medium for various intervals. Incorporation of the labeled nucleoside into nuclear DNA occurred in a fraction of the nuclei which increased with time. In addition, the cytoplasm of all cells incorporated enough tritium to be readily detected by autoradiography. The tritium was not removed by hydrolysis in 1...
